Ohio alone experiences 7,034 traffic deaths in one year due to impaired driving. Not all of the time the fatalities are just the impaired driver of the motor vehicles. A lot of the time the fatalities are of the people the impaired drivers hit while driving or the passengers of their vehicles. This problem does have an underlying solution that needs to be found and used to help the younger generations, adults, and any person affected by alcoholism in Licking County. The communities, families, friends, schools, sports teams and many more are affected by alcoholism daily because people make the decision to drive under the influence. There is a very well known program called LAPP. This is a program organized to help recovering alcoholics and drug addicts. There are several hundreds of people that attend LAPP after getting in trouble with the law or after being reported by a family member or someone they know that cares about them enough to recommend getting help.Little do people that that programs such as LAPP are a lot of the time uneffective because you cannot get a person to quit doing something that they simply do not want to quit doing. There has to be a reason or an incentive for most people that will give them a reason to give up the drinking that has lead to alcoholism. In the novel Catcher in the Rye by J.D. Salinger, Holden Caulfield, our protagonist, makes a mental journey in relation to dealing and coping with the grief accompanied with the death of his brother, and we are able to see how his mental status changes and how he needs guidance from loved ones. In this journey, he learns about how to make connections and that staying close to the people he loves can improve his outlook on life and overall mood. During the book, he isolated himself from others and always found the negative in everything, which did put him in a very grim place. In fact, he even becomes clinically depressed, as we can clearly see from our point of view. Either way, it is not their fault for their emotions and reactions to those circumstances. Emotions are uncontrollable, therefore becoming depressed or developing an xiety in reaction to a certain condition would not make it their fault. In the book, the death of Holden’s brother, Allie, really changed him. â€Å"I slept in the garage the night he died, and I broke all the goddam windows with my fist, just for the hell of it† (39). Allie’s death was an uncontrollable event, in which Holden had a very natural reaction to by punching the windows in anger. We can see that he has become very depressed and negative, and has lost the ability to form connections. This kind of behavior, from our perspective, needs some serious attention. For moments throughout the book, it almost appeared that he was considering taking his own life due to the severe depression he was experiencing. â€Å"What I really felt like, though, was committing suicide. Currently, the role of EMT in TKI drug resistance is poorly understood. EMT results in loss of tight junction proteins such as E-cadherin and upregulation of transcriptional repressors of these proteins such as ZEB1. Recently, protein arginine methyl transferase 1 (PRMT1) has been shown to be an important regulator of EMT, cancer cell migration and metastasis. However, the role of PRMT1 in TKI resistance is n ot known. In this study, we propose to evaluate the role of EMT proteins in TKI resistance using H2170 and H358 cell lines (wild type EGFR) that were made resistant to an EGFR inhibitor, erlotinib and a c-Met inhibitor, SU11274 and a combination of both. H3255 cell line (with L858R EGFR mutation), and H1975 cell line with L858R and T790M EGFR mutations which we have made resistant to erlotinib will also be used.
